Epitalon is a synthetic tetrapeptide modelled on epithalamin, a pineal gland extract developed by Vladimir Khavinson's group in St Petersburg, and is claimed to activate telomerase and act as a geroprotector. Reported effects include telomere elongation in cultured human fibroblasts and modest maximum-lifespan extension in mice, but essentially all of this work originates from one research group and has not been independently replicated.

the explanation

Epitalon is a four-amino-acid peptide from Russian research claiming to switch telomerase back on and slow ageing. Almost every study supporting it comes from the same one laboratory in St Petersburg, and no independent group anywhere else has ever reproduced the results.

regulatory status

Unapproved; no marketing authorisation anywhere

Epitalon holds no FDA, EMA or other regulatory approval, and unlike Semax it does not appear on Russia's essential medicines list; the related pineal extract epithalamin was used in Soviet-era Russian clinical research but the synthetic tetrapeptide is sold only as a research chemical. At the Pharmacy Compounding Advisory Committee meeting of 24 July 2026 the panel voted 7 to 5 with one abstention to recommend adding epitalon to the section 503A bulks list, against FDA staff advice. A recommendation is non-binding, confers no approval and is not a finding of efficacy; implementation would require notice-and-comment rulemaking. FDA has not published meeting minutes and these tallies are trade-press reporting from the room.

Epitalon is proposed to act at the level of gene regulation rather than through a conventional receptor.

Claimed telomerase induction

In telomerase-negative human fetal fibroblasts, Epitalon was reported to induce expression of the catalytic subunit of telomerase along with enzymatic activity and telomere elongation. This single in vitro result underpins most longevity claims made for the compound.

Overcoming the Hayflick limit

The same group reported that treated somatic cells continued dividing past their normal replicative ceiling. This has never been reproduced by an independent laboratory.

Pineal and melatonin signalling

Epitalon derives from epithalamin, a pineal gland extract, and is proposed to restore age-related decline in pineal function and melatonin rhythm. The mechanistic link between the synthetic tetrapeptide and pineal physiology is asserted more than demonstrated.

Genome stability in rodents

Treated SHR mice showed a 17.1% reduction in chromosome aberrations in bone marrow cells and a 6.0-fold lower incidence of leukaemia. Total spontaneous tumour incidence, however, was unchanged.

⚠ the catch

The Khavinson lifespan claims have no Western replication — in more than twenty years since the key 2003 telomerase and mouse-lifespan papers, no independent laboratory outside the originating St Petersburg group has reproduced telomere elongation, telomerase induction, or lifespan extension with this tetrapeptide. Even taken at face value the mouse data showed no change in mean lifespan and no reduction in total tumour incidence, which is a considerably weaker result than the popular framing suggests.
